Membrane sialic acid and the mechanism of insulin action in adipose tissue cells. Effects of digestion with neuraminidase.

چکیده
Mild digestion of isolated adipose tissue cells with Closfridium perjringens neuraminidase (10 to 20 mpg of purified enzyme per ml, 15 min at 37”) results in enhanced glucose transport by these cells. This effect appears to mimic closely the physiological effect of insulin on glucose transport by normal cells, and there is evidence to suggest that these two apparently different stimuli may affect similar membrane structures. Digestion of fat cells with higher concentrations of neuraminidase (about 1 pg of purified enzyme per ml, 15 min, 37”) causes a fall in the enhanced basal rate of glucose transport described above and abolishes the effects of insulin on glucose transport and on lipolysis. Such enzymatic digestion, however, has no effect on the total quantity of specific cell membrane receptor for insulin or in the affinity of the insulinreceptor complex. The biological and the receptor-binding properties of insulin are thus dissociated. The loss of insulin-stimulated glucose transport caused by neuraminidase digestion does not reverse spontaneously by prolonged incubation of the cells after digestion. Neuraminidases from C. perjringens, Vibrio cholerae and influenza virus were purified by affinity chromatography using selective agarose adsorbents containing an oxamic acid derivative. The three enzymes, at low concentration, can destroy the glucose transport effects of insulin on fat cells. However, the specific patterns of digestion observed with the three different enzymes are not identical, probably as a result of the differences in substrate specificity of the enzymes. DiEerently linked sialic acid residues at the cell surface may participate differently in various membrane functions. The effects of digesting fat cells with various combinations of C. perjringens neuraminidase and trypsin on glucose oxida-

Serum Levels of Sialic Acid and Neuraminidase Activity in Cardiovascular, Diabetic and Diabetic Retinopathy Patients
Background: Sialic acid is a component of serum that is elevated in diseases such as diabetes and certain malignancies. The normal range of SSA concentration and serum neuraminidase activity in different populations are varied, probably due to racial differences. Objective: The purpose of the present study was to obtain the average SSA concentration and serum neuraminidase activity, in an Iran...
متن کاملSimultaneous Effects of Metformin and Sitagliptin on the Contents of Insulin Resistance Proteins Glucose Transporter 4 and Protein Kinase B in Diabetic Patients\' Adipose Tissue
Objective: Obesity is a factor in the development of insulin resistance and type 2 diabetes. Obesity contributes a wide variety of metabolic changes such as insulin resistance. The insulin signal mechanism to intra-cells occurs in insulin resistance, primarily in adipose tissue cells, which can be appropriate targets for therapeutic approaches by recognizing the proteins in this pathway. The st...

insulin has been suggested by Cuatrecasas & Illiano (197 1) on the basis of neuraminidase treatment of adipose-tissue cells. Enzymic digestion was reported to have no effect on the total quantity of insulin receptor in the membranes or on the affinity of the receptor for insulin, but abolished the effect of insulin on glucose transport and on lipolysis. As shown in Fig. 1, neuraminidase action ...
متن کاملAssessment of Sialic Acid Distribution in Mouse Epididymis
Previous studies have shown that epididymal epithelium and its secretions are critical for sperm maturation. These secretions contain many glycoconjugates with sialic acid terminal sugar. This terminal sugar by interveining in cellular interactions and masking surface receptors, has an important role in sperm maturation and protection. Moreover lectins have been employed as useful probes to det...
متن کاملComparison of Characteristics of Human Amniotic Membrane and Human Adipose Tissue Derived Mesenchymal Stem Cells
BACKGROUND Mesenchymal stem cells (MSCs) are ideal candidates for treatment of diseases. Amniotic membranes are an inexpensive source of MSCs (AM-MSC) without any donor site morbidity in cell therapy. Adipose tissue derived stem cells (ASCs) are also suitable cells for cell therapy. There is discrepancy in CD271 expression among MSCs from different sources. In this study, the characteristics...
